DNA Barcode References - WG1 session 

The endless quests for completeness and curation of reference libraries

10th March 2021, morning

DNA barcode reference libraries are of immense importance for correct identification of organisms using DNA barcoding and metabarcoding. In this session we welcome contributions that focus on the construction of reference databases and analysis of reference data: from filling the gaps and quality control of existing barcode libraries, to the analyses of species boundaries and use of integrative taxonomy in revision of classifications.

Field & Lab Protocols - WG3 session

The search for "One (or none) protocol to rule them all!"

Environmental DNA as a tool in molecular ecology resulted in promising success with detection in higher precision rates and easier field protocols compared to traditional field surveys in aquatic ecosystems. Moreover, after the enhanced progress in the high throughput sequencing technologies, bioassessment of entire communities from related habitats from single or pooled PCR also becomes available without any prior knowledge on the species inhabiting the environment. One of the main challenges in eDNA studies is that the DNA extracted from potentially inhibiting agent rich environmental samples are mostly short and degraded fragments and thus requiring protocol adjustment accordingly. This session will try to highlight outcomes of all possible scenarios and discuss scientific data related to variables such as effects of sampling, filtration, storage conditions, extraction method, primer design/using universal primers, PCR (conventional, quantitative or digital), sequencing (Sanger, pyrosequencing, high throughput).

Data Analysis & Storage - WG4 session

Revealing, comparing and storing biodiversity patterns in massive amplicon datasets

Analyses of biodiversity data from metagenomics face two main issues: one on storing and maintaining large amount of data in online repositories, and the other one on having comparable pipelines to process data. Data processing aims at revealing biodiversity patterns and bridging these patterns with tools and models in statistical ecology. The session will explore the details of the current and expected approaches (i) for storage of raw data and data processing outcomes of massive datasets compliant with FAIR principles, (ii) in the available pipelines to obtain Operational Taxonomic Units, Amplicon Sequence Variants, and other basic units of diversity from massive sequencing; showcasing examples, advantages, caveats, and pitfalls. The main aims are to foster our understanding of biodiversity patterns at different scales by producing reliable and reusable data and pipelines downstream of massive sequencing and thus, enabling cross comparisons.

Implementation Strategy & Legal Issues - WG5 session

Neglected, feared but indispensable:  standardization and tailored communication

Worldwide, the use of DNA-based monitoring is developing fast and the time has come to focus on standardisation and implementation into routine monitoring. This session welcomes experiences on how to implement these novel techniques into routine water quality or biodiversity monitoring in all kind of aquatic ecosystems. We also are looking for people sharing experiences on how to communicate to stakeholders such as monitoring agencies and regulatory bodies and how to involve them in the decision making. Finally, legal issues and opportunities or threats with regard to the use of DNA-based monitoring will be discussed during this session.

Biotic Indices & Metrics - WG2 session

New trends in bioassessment of aquatic ecosystems: from organisms to DNA-based metrics

Traditionally, the biological quality of aquatic ecosystems is assessed using selected groups of organisms that can be identified morphologically. Recent advances in high-throughput genomic approaches offered new opportunities to monitor biodiversity and assess ecological status using DNA barcoding and metabarcoding. The DNA-based tools have been used in three different ways: (1) to replace morphological identification of biological quality elements in existing biotic indices, (2) to develop new molecular indices based on morphologically inconspicuous groups of potential environmental indicators, and (3) to predict biotic indices from environmental DNA datasets using machine learning methods. During this session, we will present these different approaches and discuss their advantages and challenges in view of their future application in routine bioassessment.
